Tesamorelin is a synthetic peptide that belongs to the growth hormone-releasing hormone family and contains the same forty-four amino acid sequence as endogenous GHRH, extended at the amino terminus by a trans-3-hexenoyl group. That small fatty acid modification protects the peptide from rapid cleavage by dipeptidyl peptidase-4, the enzyme that shortens the half-life of native GHRH to only a few minutes. Chemically the compound is produced by solid-phase peptide synthesis, purified by chromatography, and supplied as a sterile lyophilized powder for reconstitution.
Regulatory approval in the United States came in 2010, when the Food and Drug Administration cleared the peptide for the reduction of excess abdominal fat in adults with HIV infection and associated lipodystrophy. The decision rested mainly on two randomized phase 3 trials that enrolled roughly eight hundred patients and ran for twenty-six weeks. Participants receiving active drug showed substantially greater declines in visceral adipose tissue than those receiving placebo, while total body weight changed comparatively little. A reformulated presentation was later approved, and the product has remained a niche therapy rather than a general weight-loss agent.
Tesamorelin occupies a narrow position among agents that act on the growth hormone axis. Unlike growth hormone itself, which is given as replacement, it stimulates the pituitary to release the hormone in pulses, so the downstream increase in insulin-like growth factor 1 depends on intact somatotroph function. Other peptides in the same family include shorter GHRH fragments and synthetic secretagogues with different stability profiles. Several points remain unresolved, including whether the reduction in visceral fat translates into fewer cardiovascular events, what happens to metabolic markers after long-term use, and how the drug compares with lifestyle or surgical approaches.

Several related peptides act on the same receptor, including sermorelin, a shorter GHRH fragment, and modified analogs such as CJC-1295 and modified GRF(1-29) that are common in research settings rather than approved products. Tesamorelin differs from growth hormone itself in that it acts upstream, prompting the pituitary to release the hormone through physiological signaling rather than supplying it directly. Terminology in the literature distinguishes GHRH analogs, growth hormone secretagogues, and recombinant growth hormone, although popular discussion often blurs these categories together. Precise naming matters when comparing study results.
Tesamorelin is a synthetic peptide of 44 amino acids that reproduces the sequence of human growth hormone-releasing hormone (GHRH) and carries a trans-3-hexenoyl group on its N-terminal tyrosine. That small fatty-acid modification blocks cleavage by dipeptidyl peptidase-4, the enzyme that rapidly degrades native GHRH in plasma. The result is a molecule with a longer circulating half-life than the natural hormone while retaining the same receptor target. It is supplied as a lyophilized powder for reconstitution and belongs to the broader class of GHRH analogs studied for effects on pituitary growth hormone secretion.
Development work on the compound, originally designated TH9507, focused on conditions in which reduced growth hormone signaling is thought to contribute to altered body composition. The United States Food and Drug Administration approved it in 2010 for the treatment of excess visceral abdominal fat in adults with human immunodeficiency virus infection and lipodystrophy. Later research examined other populations, including adults with mild cognitive impairment, where a large trial did not meet its primary endpoints. This mixed record illustrates how a single mechanism can produce clear effects in one setting and inconclusive results in another.
Measuring the effect of a growth hormone-releasing hormone analogue requires markers that reflect pituitary output rather than the peptide itself. The two most frequently used are growth hormone and insulin-like growth factor 1. Growth hormone fluctuates sharply across the day and responds to sleep, stress, and meals, so isolated readings can be difficult to interpret. Insulin-like growth factor 1 changes more slowly and is often treated as the more stable integrated marker of axis activity.
Because growth hormone is released in pulses, single measurements can misrepresent overall secretion. Investigators sometimes use repeated sampling or overnight profiles to capture the pattern rather than a single value. Provocative testing, in which a stimulus is given and the response is tracked over time, offers another way to characterize the axis. Each approach carries trade-offs between sensitivity, burden on the participant, and the influence of non-target variables.
Clinical investigation has focused on HIV-associated lipodystrophy, a condition in which antiretroviral therapy contributes to abnormal fat distribution. Excess visceral adipose tissue accumulates in the abdomen while peripheral fat may be lost. Tesamorelin was evaluated for reducing this visceral fat depot, with trials measuring changes in abdominal fat by imaging rather than by body weight alone. The rationale rests on the known lipolytic effects of growth hormone. Effects on visceral fat are documented, while long-term outcomes regarding cardiovascular risk remain less clearly established.
Tesamorelin is a synthetic peptide analog of growth hormone-releasing hormone, composed of 44 amino acids. It was designed to retain the biological activity of the native hormone while resisting rapid enzymatic degradation. The compound is classified as a growth hormone secretagogue and belongs to the broader family of hypothalamic releasing factors. In research and clinical settings, it is studied for its ability to stimulate pituitary growth hormone release. Its structure includes a modification at the N-terminus that contributes to an extended half-life relative to native growth hormone-releasing hormone.
By July, the Centre was the only non-Nazi party that still even nominally existed; the SPD and KPD had been banned outright, while the others had been browbeaten into dissolving themselves. On 1 July, Papen and Kaas agreed that as part of the concordat, German priests would stay out of politics. As it turned out, the party dissolved on 5 July—much to the dismay of Cardinal Pacelli, who felt the party should at least have waited until after the conclusion of negotiations. The day after, the government issued a law outlawing the formation of new political parties, thereby making the NSDAP the only legally permitted party in Germany.
==== Carbon-oxygen ==== Glucose oxidase (GOX) catalyzes the oxidation of β-D-glucose to D-glucono-δ-lactone with the simultaneous reduction of enzyme-bound flavin. GOX exists as a homodimer, with each subunit binding one FAD molecule. Crystal structures show that FAD binds in a deep pocket of the enzyme near the dimer interface. Studies showed that upon replacement of FAD with 8-hydroxy-5-carba-5-deaza FAD, the stereochemistry of the reaction was determined by reacting with the re face of the flavin. During turnover, the neutral and anionic semiquinones are observed which indicates a radical mechanism.
Starting from the 1990s, major discoveries of exceptionally preserved fossils in deposits known as conservation Lagerstätten contributed to research on dinosaur soft tissues. Chiefly among these were the rocks that produced the Jehol (Early Cretaceous) and Yanliao (Mid-to-Late Jurassic) biotas of northeastern China, from which hundreds of dinosaur specimens bearing impressions of feather-like structures (both closely related to birds and otherwise, see § Origin of birds) have been described by Xing Xu and colleagues. In living reptiles and mammals, pigment-storing cellular structures known as melanosomes are partially responsible for producing colouration. Both chemical traces of melanin and characteristically shaped melanosomes have been reported from feathers and scales of Jehol and Yanliao dinosaurs, including both theropods and ornithischians. This has enabled multiple full-body reconstructions of dinosaur colouration, such as for Sinosauropteryx and Psittacosaurus by Jakob Vinther and colleagues, and similar techniques have also been extended to dinosaur fossils from other localities. (However, some researchers have also suggested that fossilized melanosomes represent bacterial remains.) Stomach contents in some Jehol and Yanliao dinosaurs closely related to birds have also provided indirect indications of diet and digestive system anatomy (e.g., crops). More concrete evidence of internal anatomy has been reported in Scipionyx from the Pietraroja Plattenkalk of Italy. It preserves portions of the intestines, colon, liver, muscles, and windpipe.

To reduce the cost to the military of maintaining stockpiles of certain pharmaceuticals, the United States Department of Defense (DoD) and the Food and Drug Administration (FDA) operate a joint initiative known as the Shelf Life Protection Program (SLEP), which evaluates the long-term effectiveness of medications stockpiled by the DoD and other government agencies. Under the program, medications are tested for safety and stability for extended periods of time in controlled storage conditions. In many cases, medications tested were found to be effective for years past their printed expiry dates; a 2006 study by the Journal of Pharmaceutical Sciences found that two-thirds of 122 medications tested through SLEP remained effective for an average of at least four additional years. In 2016, the DoD reported that the program had helped save the department US$2.1 billion on replacing stockpiled medications. While testing and stability assessment found that 88% of the lots were effective at least one year after their expiration date, with an average extension of 66 months, the additional stability period was very variable. For antibiotics studied, the mean extension for ciprofloxacin was 55 months, and for amoxicillin 23 months.
The Golden Gate assembly protocol was defined by Engler et al. 2008 to define a DNA assembly method that would give a final construct without a scar sequence, while also lacking the original restriction sites. This allows the protein to be expressed without containing unwanted protein sequences which could negatively affect protein folding or expression. By using the BsaI restriction enzyme that produces a 4 base pair overhang, up to 240 unique, non-palindromic sequences can be used for assembly. Plasmid design and assembly In Golden Gate cloning, each DNA fragment to be assembled is placed in a plasmid, flanked by inward facing BsaI restriction sites containing the programmed overhang sequences. For each DNA fragment, the 3' overhang sequence is complementary to the 5' overhang of the next downstream DNA fragment. For the first fragment, the 5' overhang is complementary to the 5' overhang of the destination plasmid, while the 3' overhang of the final fragment is complementary to the 3' overhang of the destination plasmid. Such a design allows for all DNA fragments to be assembled in a one-pot reaction (where all reactants are mixed together), with all fragments arranged in the correct sequence. Successfully assembled constructs are selected by detecting the loss of function of a screening cassette that was originally in the destination plasmid. MoClo and Golden Braid The original Golden Gate Assembly only allows for a single construct to be made in the destination vector .

== Discovery == Collagen XXIII was first identified and isolated from rat prostate carcinoma cells by Jacqueline Banyard, Lere Bao and Bruce R. Zetter in 2003. They also identified this protein in human tissue. They concluded that at the nucleotide level, human and rat collagen XXIII alpha 1 show 76% identity. Furthermore, cellular localization of collagen XXIII was determined by immunofluorescence staining, using an antibody that recognizes the carboxyl terminus of the protein. It was demonstrated that the carboxyl terminus of collagen XXIII is present on the cell surface.
=== Abuse potential === Pregabalin has potential for recreational use and abuse. It possesses a relatively wide therapeutic index and margin of safety compared to other central nervous system (CNS) depressants, and its effects may include euphoria, anxiolysis, and sedation. Although generally considered to have low abuse potential, reports describe pregabalin inducing sensations of euphoria, calmness, excitement, and a "high" comparable to that of cannabis. These effects may contribute to the development of substance dependence, and withdrawal symptoms can occur if the medication is stopped abruptly. While pregabalin and cannabis share certain effects, such as CNS depressant and orexigenic (appetite-stimulating) properties, they differ significantly in their pharmacodynamics. Cannabis is primarily an anxiogenic substance with mild psychedelic effects, whereas pregabalin is an anxiolytic agent associated with mild euphoria that is prone to rapid tolerance.

=== Ebola === In October 2015, the United States Army Medical Research Institute of Infectious Diseases (USAMRIID) announced preclinical results that remdesivir had blocked the Ebola virus in Rhesus monkeys. Travis Warren, who has been a USAMRIID principal investigator since 2007, said that the "work is a result of the continuing collaboration between USAMRIID and Gilead Sciences". The "initial screening" of the "Gilead Sciences compound library to find molecules with promising antiviral activity" was performed by scientists at the Centers for Disease Control and Prevention (CDC). As a result of this work, it was recommended that remdesivir "should be further developed as a potential treatment." Remdesivir was rapidly pushed through clinical trials due to the West African Ebola virus epidemic of 2013–2016, eventually being used in people with the disease. Preliminary results were promising; it was used in the emergency setting during the Kivu Ebola epidemic that started in 2018, along with further clinical trials, until August 2019, when Congolese health officials announced that it was significantly less effective than monoclonal antibody treatments such as ansuvimab and atoltivimab/maftivimab/odesivimab. The trials, however, established its safety profile.
=== AI Growth Zones and Compute Roadmap === Following the Action Plan, the government published its AI Growth Zones policy paper in November 2025, designating specific sites, with streamlined planning approvals and accelerated power connections. The policy aims to reduce the time to power for new data centres by up to five years and save a 500 MW data centre up to £80 million annually. In July 2025, the UK government published its UK Compute Roadmap outlining up to £2 billion for a modern public compute ecosystem and a 20-fold expansion of the AI Research Resource by 2030, alongside AI Growth Zones.
Existential – Existential psychotherapy postulates that people are largely free to choose who we are and how we interpret and interact with the world. It intends to help the client find deeper meaning in life and to accept responsibility for living. As such, it addresses fundamental issues of life, such as death, aloneness, and freedom. The therapist emphasizes the client's ability to be self-aware, freely make choices in the present, establish personal identity and social relationships, create meaning, and cope with the natural anxiety of living. Gestalt – Gestalt therapy was primarily founded by Fritz Perls in the 1950s. This therapy is perhaps best known for using techniques designed to increase self-awareness, the best-known perhaps being the "empty chair technique". Such techniques are intended to explore resistance to "authentic contact", resolve internal conflicts, and help the client complete "unfinished business". Postmodern – Postmodern psychology says that the experience of reality is a subjective construction built upon language, social context, and history, with no essential truths. Since "mental illness" and "mental health" are not recognized as objective, definable realities, the postmodern psychologist instead sees the goal of therapy strictly as something constructed by the client and therapist. Forms of postmodern psychotherapy include narrative therapy, solution-focused therapy, and coherence therapy. Transpersonal – The transpersonal perspective places a stronger focus on the spiritual facet of human experience.
==== Impact of the Tokyo Code (1993) ==== The Tokyo Code of 1993 extended the provisions for conserving names to all species, not just those of major economic importance. This change in the International Code of Botanical Nomenclature allowed for the conservation of names that would promote nomenclatural stability. Despite this provision, no formal proposal was made to conserve the name Parmotrema chinense, and thus it did not gain widespread acceptance. David Hawksworth's 2004 study brought significant clarity to the taxonomic confusion. He rediscovered Osbeck's original material in Linnaeus' herbarium and identified it as belonging to Parmotrema tinctorum, not Parmotrema perlatum. Hawksworth demonstrated that Lichen chinensis was not validly published because it lacked a proper description and was linked with an expression of doubt by Osbeck. Hawksworth's work led to the reinstatement of the name Parmotrema perlatum, confirming that Hudson's name was legitimate and should continue to be used. This resolution was based on the original typification by Hale and the invalid publication status of Lichen chinensis. Recent studies suggest that the circumscription of Parmotrema perlatum may need to be revised. Research utilising DNA sequencing has uncovered cryptic diversity within the genus Parmotrema, indicating that traditional phenotype-based identification methods may underestimate species diversity. Specifically, the genetic analysis of P. perlatum and related species revealed multiple distinct lineages that were previously grouped under a single nominal taxon.

It is a laboratory-made peptide of forty-four amino acids whose sequence matches human growth hormone-releasing hormone, with a modified amino terminus. The modification is a short unsaturated fatty acid chain attached to the first residue. This change slows enzymatic breakdown and lengthens the time the peptide stays active in circulation.
The clinical program was designed around HIV-associated lipodystrophy, a condition in which fat accumulates abnormally around the internal organs. Trials enrolled that specific population, so the evidence base covers it rather than the general population. Regulators approved the drug for the studied indication only, and promotion outside it is not permitted.
Growth hormone therapy supplies the finished hormone directly, while this peptide acts upstream and asks the pituitary to secrete its own. That difference means the response depends on a functioning pituitary and on the body's normal feedback loops. It also means the circulating hormone profile is pulsatile rather than a flat, injected level.
